Award Description: The proposed studies take advantage of recent technical advances, animal models, and alternative strategies to implement methodological changes in Aims 1 and 3 that will expedite progress towards our research goals by increasing data quality and acquisition rate. Supplements to Aim I propose implementing a cell-based assay to evaluate chemical effects on DMRT1 activity, while also monitoring compound specificity and impact on cell function and viability. Use of the assay will, therefore, reduce the level of post-screening evaluation for specificity and cell toxicity and consequently the time required for lead compound identification. Supplements to Aim 3 propose the addition of high throughput sequencing for DMRT1 binding site identification as well as enrichment of identified DMRT1 binding sites by comparing target sequences from Dmrt1+/+ and Dmrt1-/- mice, which will reveal and eliminate non-specific sequences from the final determined set of DMRT1 target genes. The combined advancements are projected to facilitate ongoing research and lead to earlier realization of the intended goals. .
