Award Description: The activities proposed in this Administrative Supplement are intended to accelerate the tempo of the scientific research described in the parent R01 grant , as it is to enable additional effort to be dedicated to objectives complementary to and emanating from the studies being performed in that grant. Execution of the proposed studies require the hiring of a Postdoctoral Associate who will undoubtedly contribute skills not currently in the laboratory and who will also acquire additional expertise that will increase the competitiveness of the PostDoc for future employment. The objectives of the proposed studies are to 1) complete quantitative and qualitative assessment of pulmonary SAA3 expression subsequent to NO2 exposure, 2) establish in vitro the mechanisms of apoSAA-induced dendritic cell stimulation and IL-1? secretion, 3) measure allergic sensitization and development of allergic asthma manifest by administration of apoSAA in wild-type, TLR2-, TLR4-, MyD88-, Nlrp3-, ASC-, and Caspase-1-deficient mice, and 4) generate and characterize transgenic mice inducibly secreting mouse SAA3 exclusively by airway epithelium.
