Award Description: This supplement is intended to accelerate the development of an AAV4-BDNF and noggin-based HD treatment strategy, by expanding our assessment of this approach to the non-human primate brain. We intend to assess the efficacy of AAV4-mediated BDNF/Noggin in the squirrel monkey brain, in terms of both the dose-response relationship between viral dose and striatal neuronal addition, and the duration of functional transgene expression achieved by the AAV4 in the primate. We will then assess the integration and anatomic relationships of the newly added striatal neurons, defining their geographic distribution, transmitter phenotypes and pallidal targets achieved, all in the adult monkey brain. These data should provide us the basis for assessing the therapeutic potential of induced neuronal addition in primate models of HD.
