Award Description: PROCUREMENT OF ADDITIONAL NEEDED EQUIPMENT (Specific Airns 1-3 of the parent R01). The assay of clinical samples using a sandwich ELISA developed in our laboratory to specifically measure the full-length forms of acyl- and des-acyl ghrelin is a key aspect of our funded clinical research studies. To speed progress of the assays we want to purchase a 384-well plate washer. These plates are required to get the necessary throughput for the large number of samples being generated in Specific Aims 1-3. This will enhance the efficiency and throughput, lower the coefficients of variation and improve the quality of the data. RELEVANCE: A critical aspect of the parent grant is the assay of clinical samples using a sandwich ELISA that can specifically measure the full-length forms of acyl- and des-acyl ghrelin. This new equipment will enhance throughput and accuracy. A model of ghrelin regulation that identifies other significant components and explains variation and inter-conversion of acyl- and desacyl ghrelin will be more physiologically relevant.
